Purpose: This kit is designed to quantify interleukin-23 (IL-23) in human serum, plasma, and other related biological samples.
Principle of Detection: The assay is based on the double-antibody sandwich ELISA method. A microplate is pre-coated with a specific antibody against IL-23. After adding the sample, IL-23 binds to the immobilized antibody. An HRP-conjugated detection antibody is then added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following washing steps, TMB substrate is added, which turns blue under HRP catalysis and changes to yellow upon acid termination. The intensity of the color is directly proportional to the IL-23 concentration in the sample. Absorbance is measured at 450 nm using a microplate reader, and concentrations are calculated from a standard curve.

Sample Requirements:
Samples should be processed as soon as possible after collection. If not tested immediately, store at -20°C, avoiding repeated freeze-thaw cycles. Avoid using samples containing NaN3, as it may inhibit HRP activity.
Procedure:
1. Prepare standards by diluting the original standard according to the provided chart.
2. Add 50 μl of standard or sample diluent to each well, followed by 10 μl of the sample (final 5-fold dilution).
3. Seal the plate and incubate at 37°C for 30 minutes.
4. Wash the plate 5 times using 20× diluted washing solution.
5. Add 50 μl of enzyme-labeled reagent to each well except blank.
6. Incubate again for 30 minutes at 37°C.
7. Wash the plate again.
8. Add 50 μl of Color Reagent A and 50 μl of Color Reagent B, mix gently, and incubate at 37°C for 15 minutes.
9. Add 50 μl of stop solution to each well to terminate the reaction.
10. Measure absorbance at 450 nm within 15 minutes of stopping the reaction.
