Activated adsorbent
The solid phase extraction cartridge is rinsed with a suitable solvent prior to sample extraction to keep the adsorbent moist and to adsorb the target compound or interfere with the compound. Different modes of solid phase extraction cartridge activation are different with solvents.
1 A weakly polar or non-polar adsorbent for reverse phase solid phase extraction, usually rinsed with a water-soluble organic solvent such as methanol, and then rinsed with water or a buffer solution. It can also be rinsed with a strong solvent (such as hexane) prior to rinsing with methanol to eliminate impurities adsorbed on the adsorbent and its interference with the target compound.
2 The polar adsorbent used in the normal phase solid phase extraction is usually rinsed with the organic solvent (sample matrix) in which the target compound is located.
3 The adsorbent used in ion exchange solid phase extraction can be rinsed with a sample solvent when used in a sample in a non-polar organic solvent; when it is used in a sample in a polar solvent, it can be rinsed with a water-soluble organic solvent. Then, it is rinsed with an appropriate pH and an aqueous solution containing a certain organic solvent and salt.
In order to keep the adsorbent in the solid phase extraction cartridge moist until it is added to the sample after activation, approximately 1 ml of the solvent for the activation treatment should be maintained on the adsorbent after the activation treatment.
2. Loading
The liquid or dissolved solid sample is poured into the activated solid phase extraction cartridge, and then the sample is introduced into the adsorbent by vacuuming and pressurizing.
3. Washing and elution
After the sample enters the adsorbent and the target compound is adsorbed, the weakly retained interfering compound can be washed away with a weaker solvent, and then the target compound is eluted with a stronger solvent and collected. Leaching and elution As described above, the eluent or eluent can be passed through the adsorbent by vacuuming, adding or centrifuging.
If the adsorbent which adsorbs the target compound is weak or non-adsorbed and has strong adsorption to the interfering compound, the target compound can be first rinsed and collected to retain the interfering compound (adsorption). On the adsorbent, the two are separated. In most cases, the target compound is retained on the adsorbent and finally eluted with a strong solvent, which is more conducive to the purification of the sample.
