First, How to Use the Instrument

Third, Working Principle
In the soil-plant-atmosphere continuum system, roots continuously absorb water from the soil, while leaves transpire moisture into the atmosphere. This creates a water potential gradient that drives water movement through the root, stem, and leaf. The xylem vessels experience negative pressure due to the low water potential in the air, which generates a pulling force that helps transport water upward.
When a branch or leaf is cut, the tensioned water column within the xylem breaks, causing the water to retract into the slit. The cut sample is then placed into the pressure chamber (as described in the operating procedure). One end of the slit extends out of the chamber, which is sealed. Pressure is applied until the water column is re-established at the cut surface, and the pressure required to do so represents the plant’s water potential.
