Working Principle:
In the soil-plant-atmosphere continuum, roots absorb water from the soil while leaves transpire, creating a continuous flow of moisture. This process generates a water potential gradient within the plant’s root, stem, and leaf system. The xylem vessels contain small water columns that are pulled upward due to the negative pressure created by the air's low water potential.
When a branch or leaf is cut, the tensioned water column breaks, causing the water to contract into the slit. The cut sample is then placed into the pressure chamber. By applying pressure, the water column is re-established at the cut surface, and the pressure required to do so corresponds to the plant's water potential.
