Previous studies have shown that the widespread use of extra virgin olive oil in the Mediterranean diet has many health benefits. To test the relationship between extra virgin olive oil and dementia, Dr. Praticò and his colleagues used a mouse model of Alzheimer's disease. The researchers divided the animals into two groups, one given a diet rich in extra virgin olive oil and the other receiving a regular diet without extra virgin olive oil. When the mice were 6 months old, symptoms of Alzheimer's disease began to appear before the introduction of olive oil into the diet.

The study found that mice fed a diet rich in extra virgin olive oil performed better at tests of working memory, spatial memory and learning ability at 9 and 12 months of age. In addition, brain tissue from both groups of mice showed significant differences in the appearance and function of nerve cells.

Dr. Praticò said: “The most obvious difference is the integrity of the synapse. The synaptic integrity of mice fed a diet rich in extra virgin olive oil is better. Dr. Praticò explained: “This is an exciting finding for us. Since autophagy is activated, memory and synaptic integrity are preserved, and the pathological symptoms of Alzheimer's disease model mice are significantly reduced. Dr. Praticò and colleagues plan to add extra virgin olive oil to the diet at the age of 12 months in the same mouse, when they have beta-amyloid plaques and neurofibrillary tangles. Dr. Praticò added: “The disease usually occurs when a patient is suspected of having Alzheimer's disease and goes to the doctor. We want to know if adding olive oil to the diet after the disease can stop or reverse Alzheimer's disease.”
