It typically contains sodium chloride, potassium chloride, sodium bicarbonate, phosphate buffers, glucose, calcium, magnesium, and phenol red as a pH indicator. The solution can be modified based on specific experimental needs, such as calcium- or magnesium-free versions, or with the addition of HEPES for enhanced buffering capacity.
HBSS serves as a base for preparing complete cell culture media and is commonly used to dilute concentrated amino acid or vitamin solutions. It is also ideal for washing cells, rinsing tissues, or preparing cell dissociation solutions like trypsin-EDTA. The 5× concentrate must be diluted to 1× before use, typically by mixing 1 part HBSS with 4 parts sterile water or deionized water.
This product is manufactured using ultrapure water, has a pH of 7.4, and is sterilized through a strict filtration process to ensure low endotoxin levels. It is stored at 4°C and has a shelf life of 3 months. When handling this solution, it is important to follow aseptic techniques to prevent contamination. Due to its high salt content, minor precipitation may occur, which can often be resolved by warming the solution. If the precipitate does not dissolve, the product should be discarded. Always wear lab Gloves and a lab coat for safety during use.
